A wide variety of cell types and growth factors modulate both the expression and activity of TGF-beta. In part, these effects are mediated by TGF-beta binding to a number of serum and extracellular matrix (ECM) proteins. TGF-beta type I receptor (TbetaRI) binds with high affinity to all three TGF-beta isoforms. To determine whether ECM binding is regulated by tyrosine sulfation of TbetaRI, we studied sulfation of TbetaRI in cell-free systems and in cell-free and cell-based binding assays. In cell-free systems, the sulfation of TbetaRI did not alter binding to heparin, vitronectin, or laminin. However, increasing sulfation of TbetaRI reduced its ability to bind to collagen. This effect was most pronounced when TbetaRI was sulfated with either a low-density sulfation-specific tyrosine sulfotransferase (SULT2B1b) or a high-density sulfation-specific tyrosine sulfotransferase (SULT2B1). Two TGF-beta isoforms, TGF-beta1 and TGF-beta3, differ at position 212, a position that has been shown to be critical for heparin binding.
